Optimasi komposisi media untuk mikropropagasi tanaman kupa (Syzygium polycephalum (Miq.) Merr. & L.M Perry)

Kupa (Syzygium polycephalum (Miq.) Merr. & L.M Perry) merupakan tanaman berkayu yang dapat dijadikan sebagai bahan obat dan zat pewarna. sudah jarang ditemukan di beberapa wilayah Indonesia, oleh karena itu diperlukan upaya konservasi. Kultur jaringan adalah salah satu teknik diaplikasikan untuk perbanyakan Penelitian ini bertujuan mendapatkan komposisi media optimum mikropropagasi kupa. terdiri atas 3 tahap percobaan, yaitu multiplikasi tunas menggunakan eksplan buku tunggal pucuk pada dasar DKW MS, mengandung BAP IBA; perakaran substrat agar atau campuran pasir + vermikulit, ditambah larutan aklimatisasi. Semua percobaan Rancangan Acak Lengkap Faktorial. Hasil penelitian menunjukkan bahwa MS 1 mg L-1 IBA 0,1 terbaik tinggi dari tunggal. Tunas terbanyak didapatkan L-1. Media Pasir Vermikulit dengan maupun memberikan 100% berakar. Planlet berasal vermikulit pemberian menghasilkan jumlah panjang akar lebih baik, daya hidup tertinggi aklimatisasi.ABSTRACTKupa is a woody plant, which has many benefits including herbal medicine and coloring matter. rarely found in some parts of therefore the conservation needed. Tissue culture technique applied for propagation conservation. This study aimed to obtain optimal composition kupa micropropagation. The consisted steps, i.e. shoot multiplication using single node tips explants cultured basic containing rooting two different substrates, mixture vermiculite sand, enriched with or solution, acclimatization. All experiments used factorial completely randomized design. results showed that +BAP 0.1 was best height from explants. L-1+ multi-shoots tip explant. Sand solution produced rooted-plantlets. Plantlets derived sand gave root number length, it also highest survival rate at acclimatization stage.
